Context
This line is spoken by Pam De Beaufort, played by Kristin Bauer van Straten, in the TV show True Blood (2008-2014).
Pam is the co-owner of Fangtasia in True Blood. Think your local sports bar, but with a lot more of the red stuff, except it's blood instead of wine. She might wear pastel colors and crack jokes occasionally, but Pam is still a fierce vampire who only really cares about Eric, the guy who made her a vampire over a century ago.
When she says this line, Pam is threatening Lafayette (played by Nelson Ellis), who needs to her sell all the vampire blood he is carrying or things could get violent. Vampire blood is the addictive substance of choice in the True Blood universe, and a big part of Fangtasia's profits, so she definitely means business.
